We are living in Hong Kong and have US passports. We want to go to Shenzhen on Friday. Can we get Visa's at the border or do we have to get them before we leave? If we have to get them in Hong Kong what is the quickest way to do that?

You do indeed need a visa for ShenZhen or any where else in "mainland" China for that matter.
There are many visa agents in MongKok that process within 24 hrs and should run you around 3500 HKD. 72 hrs should run around 1800 HKD.
It will be a single entry - 30 day tourist/visit visa.
