travel to Cuenca

I have looked for answers here and still have a few questions. Any help is appreciated. We are looking to travel to Cuenca August 21-28 to see if this is even an option for us to consider moving to from the US. We will land in Guayaquil. Questions:
1) how long and what is the cost for a van from Guayaquil to Cuenca?
2) Are there recommendations that expats use for this ride?
3) Should we buy a phone once in Cuenca?
4) Should we hire a tour guide?
5) What itinerary did you use when visiting for the first time?

We have not travelled in Latin America at all, just Mexico. Our Spanish is, well, slime to none. (we will take lessons if we decide to move).
